Testing Principle
The complex formed by the haematein-oxidised form of haematoxylin by potassium iodate – with potassium aluminium sulphate is the active chemical species of Gill’s Haematoxylin III.
The positive charge of the complex enables chemical binding to the anionic sites in the histone proteins of chromatin, causing the nuclei to take on a blue-violet colour.
