Testing Principle
The Giema stain is a complex mix composed of methylene blue and azure that can bind to the nuclei and micro-organisms, turning them dark blue when viewed through the microscope. This solution increases the intensity of the nuclear staining and can selectively highlight the cell elements.
It can be used in combination with the May Grünwald acc. to Pappenheim solution, which consists of methylene blue eosinate and which stains the nuclei blue and the basophilic cytoplasm pink-red, with the granules taking on a stain ranging from red to purple, depending on their nature, that can be acidophilic or basophilic.
